2,3,4-Trifluoroaniline — 234TFA — is a colourless to amber liquid carrying three fluorines on consecutive ring carbons next to an amino group. It is obtained by reducing 2,3,4-trifluoronitrobenzene.

Three fluorines on one ring produce a strongly electron-poor, weakly basic amine that resists metabolic oxidation almost completely. That combination of stability and electronic character is what medicinal chemists are buying, particularly in the fluoroquinolone antibacterial families.

Aromatic amines are absorbed through intact skin and can affect the blood by forming methaemoglobin. Chemical-resistant gloves, coveralls, and eye protection are required.

What is 2,3,4-Trifluoroaniline used for?

It is a pharmaceutical intermediate, used in fluoroquinolone antibacterial synthesis and other routes needing a heavily fluorinated aniline. It also serves agrochemical and specialty material chemistry.

How is 234TFA manufactured?

By reducing 2,3,4-trifluoronitrobenzene, usually by catalytic hydrogenation. The nitro compound is itself made by halex fluorination of the trichloro analogue, so the whole chain runs through three products in this range.

What do three fluorines do to the amine?

They withdraw electron density strongly, making the amine markedly less basic and less nucleophilic than aniline. Acylation and condensation therefore need more forcing conditions or more reactive reagents.

Why is heavy fluorination valuable in drug molecules?

Fluorine blocks the ring positions where metabolic oxidation would normally occur, raises lipophilicity, and shifts electron distribution. Together those effects can substantially improve a compound's stability and activity.

Are the fluorines reactive?

Not once the nitro group has been reduced. Without that activation the carbon–fluorine bonds are very stable, so the amine is the reactive handle and the fluorines remain in the final molecule.

Why does the liquid darken during storage?

Aromatic amines oxidise on contact with air and light. Nitrogen blanketing and cool, dark storage slow this considerably, and moderate colour development does not usually affect assay.

Is it soluble in water?

Only slightly. It is readily soluble in alcohols, ethers, esters, and aromatic hydrocarbons, so reactions are run in organic media.

What purity do you supply?

Standard grade is supplied at a minimum GC assay of 99% with isomer content at or below 0.5% and water at or below 0.2%. Tighter specifications can be manufactured for regulated pharmaceutical intermediate supply.
