What is 2,6-Difluoroaniline?

2,6-Difluoroaniline — 26DFA — is a colourless to pale amber liquid with fluorine atoms on both carbons flanking the amino group.

Two ortho fluorines pull hard on the nitrogen's electron density, making this one of the least basic simple anilines available. The amine is correspondingly harder to acylate, but the resulting amides carry a fluorinated, metabolically robust ring that is valuable in both drug and crop-protection design.

Handling precautions

26DFA is a toxic aromatic amine, harmful in contact with skin, by inhalation, and if swallowed. It is for industrial use by trained personnel only.

Aromatic amines are absorbed through intact skin and can affect the blood by forming methaemoglobin. Chemical-resistant gloves, coveralls, and eye protection are required.

The product is combustible. Keep away from ignition sources and earth equipment during transfer.

What is 2,6-Difluoroaniline used for?

It is an intermediate for fluorinated pharmaceuticals and agrochemicals, used where a doubly fluorinated aniline fragment is needed in an amide, urea, or heterocyclic structure.

Why are two ortho fluorines useful?

They block both positions next to the nitrogen against metabolic oxidation and unwanted substitution, and they lower the basicity of the amine substantially. The result is a robust fluorinated fragment that survives in the final molecule.

Does the low basicity make it harder to work with?

It does affect conditions. Acylation typically needs a more reactive acylating agent or more forcing conditions than a plain aniline would require, so routes are usually designed around that rather than adapted from unfluorinated chemistry.

Are the fluorines reactive?

Not under ordinary conditions. Without strong activating groups on the ring, the carbon–fluorine bonds are very stable, so the amine is the reactive handle.

How does 26DFA differ from 2,6-dichloroaniline?

Both are doubly ortho-substituted anilines, but fluorine is much smaller than chlorine. The chloro compound is dominated by steric hindrance and twisted amide geometry; the fluoro compound is dominated by electronic effects and metabolic stability.

Why does the liquid darken during storage?

Aromatic amines oxidise on contact with air and light. Nitrogen blanketing and cool, dark storage slow this considerably, and assay is usually unaffected by moderate colour development.

Is it soluble in water?

Only slightly. It is readily soluble in alcohols, ethers, esters, and aromatic hydrocarbons, so reactions are run in organic media.

What purity do you supply?

Standard grade is supplied at a minimum GC assay of 99% with isomer content at or below 0.5% and water at or below 0.2%. Tighter specifications can be manufactured for pharmaceutical intermediate use.
