What is Ortho Fluoro Aniline?

Ortho Fluoro Aniline — OFA, or 2-fluoroaniline — is a colourless to amber liquid carrying a fluorine atom on the carbon next to the amino group.

Placing fluorine ortho rather than para changes the molecule's behaviour meaningfully. The amine becomes less basic, and the fluorine sits close enough to participate when the nitrogen is used to close a ring, which is why the ortho isomer is favoured in benzoxazole, benzimidazole, and quinoline synthesis.

Storage and guidelines

Handling precautions

OFA is a toxic aromatic amine, harmful in contact with skin, by inhalation, and if swallowed. It is for industrial use by trained personnel only.

Aromatic amines are readily absorbed through intact skin and can affect the blood by forming methaemoglobin. Chemical-resistant gloves, coveralls, and eye protection are required.

The product is combustible above its flash point of about 66°C. Keep away from ignition sources and earth equipment during transfer.

Use local exhaust ventilation for drum decanting, charging, and heated operations; avoid breathing vapour or mist.

What is Ortho Fluoro Aniline used for?

It is an intermediate for fluorinated pharmaceuticals and agrochemicals, and particularly for heterocycle formation where the ortho position needs to be available for ring closure.

How does OFA differ from PFA?

Both are C6H6FN with a weight of 111.12, differing only in whether the fluorine sits ortho or para to the amino group. The ortho isomer is less basic and is chosen for cyclisation chemistry; the para isomer is the more general-purpose building block.

Why is the ortho position significant?

Reactions that close a ring onto the aniline need the position next to the nitrogen to be involved. With fluorine there, that position is both blocked to unwanted substitution and available as a leaving group under activation, which shapes what heterocycles can be made.

Is the fluorine reactive?

Not under ordinary conditions. On a ring without strong activating groups, the carbon–fluorine bond is very stable, so the amine is the reactive handle in most syntheses.

Why does the liquid turn amber or brown?

Aromatic amines oxidise slowly on contact with air and light. Assay is usually unaffected, but sealed, cool, dark, nitrogen-blanketed storage preserves colour for pharmaceutical intermediate use.

Is it soluble in water?

Only slightly. It is readily soluble in alcohols, ethers, esters, and aromatic hydrocarbons, so reactions are normally run in organic media.

What purity do you supply?

Standard grade is supplied at a minimum GC assay of 99.5% with water content at or below 0.2%. Higher assay and low-colour specifications for pharmaceutical intermediate use can be manufactured to order.

How long can it be stored?

12 months from manufacture in sealed original packaging under the recommended conditions. Nitrogen blanketing and cool, dark storage materially extend colour stability.
